Benefits of Single-Use Transfer Lines in Biopharmaceutical Applications

To meet existing and future demand and maintain a competitive advantage, many biopharmaceutical manufacturers are striving to develop increased efficiencies within manufacturing processes. This is where single-use systems, particularly single-use transfer lines, can deliver significant value, including added flexibility, improved production yields and increased cost savings.

One application for single-use transfer lines is in the aseptic transfer of inoculums through a seed train of stainless steel bioreactors. A traditional facility will feature a series of bioreactors that increase sequentially in capacity and are connected by CIP/SIP piping. Replacing the fixed piping with single-use transfer lines eliminates the need for CIP validation and reduces maintenance and capital expense by eliminating expensive valve and sanitary piping assemblies. Using Colder Products Corporation Steam-Thru® Connections allow operators to independently steam-on both ends of the transfer lines when sterilizing each bioreactor. Transitioning the SIP valves to the open position allows aseptic transfer of inoculums.

The benefits of single-use transfer lines also apply to suite-to-suite transfers. Most bioprocessing facilities have separate suites dedicated for distinct processes e.g. fermentation, harvest clarification, chromatography or formulation. Traditionally the processes have been linked together via stainless steel piping; now manufacturers can simply run a single-use transfer line through a transport door to quickly and easily link one suite to another. In the final fill process a single-use transfer line can deliver sterile drug liquid from a stainless steel holding tank to a filling machine. In these installations, the transfer line incorporate Steam-Thru connection attached to the sterile holding tank, a “through-the-wall” fluid transfer system to bring a portion of the transfer line into an isolator where filling occurs.

New Optical O2 sensors from METTLER-TOLEDO Ingold

The InPro 6880i series is a new line of highly innovative optical sensors for the measurement of Dissolved Oxygen in cell culture, fermentation and chemical processing. Based on a chemo-optical measurement principle, the InPro 6880i sensor provides significant advantages for the BioPharm industry. Extremely quick start up and response time, combined with the excellent measurement quality of this revolutionary system, make it the ideal choice for cell culture, fermentation and other BioPharm processes.

The new InPro 6880i can be sterilized and installed in the bioreactor and fermenter as before, through sensor housings mounted on Ingold ports, but the InPro 6880i optical sensor does not need electrolyte or polarization eliminating inadvertent handling errors and production delays!

Inside the InPro 6880i a green-based LED light emanating from sensor electronics excites marker molecules immobilized in an oxygen-sensitive membrane layer. The excited molecules emit red light through a chemo-optical fluorescence process. An electronic detector determines the oxygen content based upon the characteristics of the fluorescent light produced.

This innovative technology, combined with Mettler-Toledo INGOLDs long-standing excellence in the measurement of oxygen in bioreactors, was refined to produce the most advanced measurement system for oxygen monitoring applications. The result: a new standard of excellence for enhanced performance and ease of use. The InPro 6880i produces extremely reproducible results with outstanding accuracy, leading to high measurement consistency.

In addition, the InPro 6880i contains the Mettler-Toledo’s groundbreaking ISM (Intelligent Sensor Management) technology, which helps to increase the safety and accuracy of the DO measurement through on-going real time sensor diagnostics. Sensor status information is analyzed online, providing operators with real-time warnings when sensor aging signs appear.

ISM also provides a plug-n-measure functionality with automatic uploading of sensor calibration data to the system’s transmitter.
